چکیده

sequencing of a part of the amelogenin gene. This gene is lecated on the X and Y chromosomes, and there are 54 nucleotide de]etions on the Y chromosome-specific gene. Forty-seven (26 ma]e and 21 female) DNA samples and 23 (13 ma}e and 1O female) DNA samples, respectively extracted from white blood cells and hairs ofJapanese black bears were analyzed, The primers SE47 and SE48 from this X-Y homologous region were used in sex identification by PCR arnplification. These primers amplified Xand Y-specific bands, which could be used to discrirninate between sexes by a length polymorphism in al] samp]es. We suggest that PCR amplifieation using the primers SE47 and SE48 is usefu1 for sex delermination of the Japanese black bear and could be applied to DNA analysis of small samples such as hairs. Amelogenin Gene Failure in Sex Determination of Dry Human Teeth Specimens from Sokoto, Northwestern Nigeria

The study of teeth reveals a lot concerning forensic medicine. Particularly it is useful in human identification. In addition to determination of age, sex can also be determined from teeth. The genetic difference between males and females is defined by the presence or absence of the Y-chromosome. The majority of the DNA of the sex chromosomes is specific to either the X or Y form. There are reg...

Polymerase Chain Reaction (PCR)-Based Sex Determination Using Unembalmed Human Cadaveric Skeletal Fragments From Sokoto, Northwestern Nigeria

The strategy developed for sex determination in skeletal remains is to amplify the highly degraded DNA, by use of primers that span short DNA fragments. To determine sex of unembalmed human cadaveric skeletal fragments from Sokoto, North-western Nigeria, using Polymerase Chain Reaction (PCR). A single blind study of Polymerase Chain Reaction (PCR)-based sex determination using amelogenin gene a...

Prenatal sex determination in suspicious cases of X-linked recessive diseases by the amelogenin gene

Objective(s):To determine the fetal discernment in suspected cases of sex linked recessive disease in the first trimester of pregnancy. Materials and Methods: After collection of 100 Chorionic Villi samples, the DNAs were extracted and baby gender was determined. Meanwhile, after increasing the sensitivity, the system was able to detect the sex of each cell which was obtained by biopsy. Resul...

A sexing protocol for wild ruminants based on PCR amplification of amelogenin genes AMELX and AMELY (short communication)

Based on the sequences of the bovine amelogenin genes, we have designed a protocol for sexing DNA samples of wild ruminants. Basically the protocol consists on the co-amplification of two specific fragments, one from Ychromosome and one for the X chromosome, making the use of a PCR control unnecessary. It has been demonstrated to be useful for sex identification in a total of 164 samples belong...

Sex identification based on AMEL gene PCR amplification from blue sheep (Pseudois nayaur) fecal DNA samples.

The use of noninvasive genetic sampling to identify the sex of wild animals is an extremely valuable and important tool in molecular ecology and wildlife conservation. Sex determination using the amelogenin gene has been conducted in many species because only a single pair of primers is required to amplify both X- and Y-linked alleles. However, this method has not been used in field research wi...
